Abstract

Glucocorticosteroids are compounds utilized for their anti-inflammatory properties in racing animals. This research describes a liquid chromatography multiple mass spectrometry (LC/MSn) method for characterizing and quantifying 24 glucocorticoids in equine urine and plasma and in canine urine. This atmospheric pressure chemical ionization (APCI) procedure follows a liquid/liquid extraction using 1 ml of sample and 5 ml of methyl tert-butyl ether.;The method evaluation involved utilizing spiked calibration samples in equine urine and plasma and in canine urine. This method was applied to urine elimination studies incorporating dexamethasone and fludrocortisone. The fludrocortisone study produced a metabolite with m/z of 384. The molecular formula for the metabolite tentatively named 9-alpha-fluoro-3, 11-beta, 17-alpha, 20, 21-pentahydroxy-4-pregnene is C21H33FO5.
